Wragg, June B. (Agricultural Research Service, Beltsville, Md.), Howard Reynolds, and Michael J. Pelczar, Jr. Free amino acids in serine-antagonized cells of Tetrahymena pyriformis. J. Bacteriol. 90:748-754. 1965.-Growth inhibition of Tetrahymena pyriformis by l-serine in a chemically defined medium was reversed by l-arginine in a manner which resembled competitive antagonism. Composition of the free amino acid pools from cells grown in either a balanced amino acid mixture or a mixture with serine concentrations which inhibited growth suggested an antagonism by serine with energy-yielding reactions. Growth in media with excess serine resulted in the accumulation of higher concentrations of free cellular amino acids and an apparent increase in the rate of conversion of arginine to ornithine, as compared with growth in the balanced medium. The results suggested that serine or a metabolic product of serine interferes with the formation of pyruvic acid. In the presence of high levels of serine, arginine appeared to be metabolized more rapidly and to be spared when alanine, aspartic acid, or glutamic acid was added to the unbalanced medium.

Nalidixic acid and macromolecular metabolism in Tetrahymena pyriformis: effects on protein synthesis.
A study on the effect of nalidixic acid on macromolecular metabolism, particularly of protein, in Tetrahymena pyriformis was performed. It was shown that the compound is a potent inhibitor of deoxyribonucleic acid, ribonucleic acid, and protein synthesis for this organism. A conspicuous breakdown of polysomes, accompanied by the accumulation of 80S ribosomes, occurred in cells incubated for 10 ...
متن کاملFree and nonprotein amino acids of Tetrahymena pyriformis.
In a previous study (l), the composition of total cellular amino acids of the ciliated protozoan, Tetrahymena pyriformis (formerly, T. geleii), was presented together with other pertinent data concerning the nitrogen metabolism of this organism. The present investigation was undertaken to obtain information on the free and nonprotein amino acids of this protozoan cell, inasmuch as the nonprotei...
متن کامل15N investigation into the effect of a pollutant on the nitrogen metabolism of Tetrahymena pyriformis as a model for environmental medical research.
A pilot study was performed to examine the potential of stable isotope techniques for monitoring the impact of a harmful substance on the cellular nitrogen metabolism in the ciliate species Tetrahymena pyriformis. After identical cultivation periods of control cells and toluene-exposed cells in a defined culture medium enriched with [guanidino-15N2]l-arginine, a number of nitrogen-containing po...
متن کاملRegulation of cellular osmolarity and volume in Tetrahymena.
Freshwater Protozoa are hyperosmotic to their environment. Direct determinations of intracellular osmolarity on Amoeba proteus and Pelomyxa carolinensis have given values between 101 and 117 m-osmole/kg cells, when the external osmolarity was 2-8 m-osmole/1 (Lovtrup & Pigon, 1951; Riddick, 1968; Schmidt-Nielsen & Schrauger, 1963). No measurements were made of intracellular osmolarity in respons...
متن کاملThe metabolism of cyclopropane fatty acids by Tetrahymena pyriformis.
Cyclopropane fatty acids are readily metabolized by whole cells of Tetrahymena pyriformis when the fatty acids are presented either as components of intact Escherichia coli cells or as free fatty acids added to the medium. cis-ll,lZ[methylene-14C]Methyleneoctadecanoic acid was degraded with the production of 14COz by whole T. pyriformis cells or a particulate fraction derived from them. In the ...
